For random-primed DNA labeling with digoxigenin-dUTP, alkali-labile and detection of hybrids by enzyme immunoassay

Random-primed labeling of DNA probes with DIG-11-dUTP, alkali-labile and color detection of DIG-labeled hybrids. DIG-labeled probes are used for hybridization in Southern, northern, and dot blots, as well as for colony and plaque screening.
DIG-labeled hybrids are detected with an anti-DIG-alkaline phosphatase conjugate and the substrates NBT (nitroblue tetrazolium salt) and BCIP (5-bromo-4-chloro-3-indolyl phosphate, toluidinium salt), which give a light-blue precipitate.

Sensitivity and specificity: The standard labeling reaction with 1 ?g template will give a yield of 260 ng DIG-labeled DNA after a 1-hour incubation at 37?C, or 780 ng after a 20-hour incubation. The sensitivity of DIG detection depends on the concentration of the DIG-labeled probe in the hybridization reaction, and the duration of the color reaction.

原理：用DIG-dUTP為底物，隨機摻入法進行DNA標記，通過酶聯(lián)免疫雜交的方法檢測。該試劑盒可以標記25次反應，每次標記的DNA量為10ug-20ug，并可做50張10×10cm膜的雜交和檢測。

應用：非放射性DNA標記及檢測試劑盒以檢測0.1pg的同源DNA，能檢測1ug哺乳動物DNA中的單拷貝基因。跟放射性標記系統(tǒng)比較，此試劑盒能快速得到實驗結果，而且消除了放射性同位素的不安全問題。從DNA標記和雜交能見到檢測結果，整個實驗過程24小時內能完成。這試劑盒的靈敏度與特異性使它適用于所有的雜交技術以替代放射性同位素標記和放射自顯影術。這個標記方法和檢測系統(tǒng)適用于DNA—印跡轉移、菌落雜交、噬斑雜交及原位雜交。

Application

This kit was assembled with convenience in mind, offering a ready-made blocking solution, combined stock solution of NBT/ BCIP, and DIG Easy Hyb granules. The DIG-High Prime mixture includes stabilized Klenow enzyme, nucleotides, primers, and reaction buffer, all in one convenient reagent.
 
Benefits
The High Prime method of labeling offers many benefits. Plasmid, cosmid, and lambda phage DNA have all been used as templates. We have achieved equal efficiency when labeling 10 ng to 3 μg of template DNA, which may be linear or supercoiled.

The DIG-High Prime mixture includes stabilized Klenow enzyme, nucleotides, primers, and reaction buffer, all in one convenient reagent.

Assay Principle: 高效DNA地高辛標記和檢測試劑盒II是用地高辛（地高辛：英文名Digoxigenin ，簡稱DIG；又稱異羥基洋地黃毒甙元，是一種類固醇半抗原分子）標記的DNA探針，應用于分子雜交和隨后的化學發(fā)光檢測。羅氏公司的該試劑盒檢測主要分三階段進行：
1. DNA標記：根據(jù)隨機引物標記技術，生成DIG地高辛標記的DNA探針。
2. 雜交：按照標準雜交方法進行，將地高辛標記的DNA探針用于核酸點雜交，可選擇帶正電的尼龍膜或純硝酸纖維素膜作為雜交膜。
